【要旨】Marburg virus is a highly pathogenic virus causing hemorrhagic fever in humans with high mortality rates. Structural investigations into the virus have been limited both by the need to prepare samples in Biosafety level 4 facilities and also by the pleomorphic nature of the virus. In this work, we present a detailed investigation, by a combination of cryo-transmission electron microscopy, cryo-electron tomography and subtomogram averaging, of the structure of the nucleocapsid within intact virions. We also examine the structure of the nucleocapsid during the budding process in infected cells and demonstrate that there is an absolute directionality to this process. Initial findings on the structure of the matrix in intact virions and in a VLP system will be presented.
